چکیده

Two iron-sulfur proteins from Bacillus polymyxa (ferredoxins I and II) have been shown to transfer 1 electron per 4 iron atoms at a potential of -380 and -420 mv, respectively, at pH 7. The proteins contain 4 iron and 4 labile sulfur atoms and 4 cysteine residues in molecules of about 9,000 daltons; electron paramagnetic resonance and optical properties suggest that each contains an iron-sulfur cluster similar to one of those present in bacterial-type ferredoxins which contain 8 iron atoms distributed between two clusters. These proteins therefore represent an example of a subclass of the bacterial-type ferredoxins, in which 4 iron atoms appear to occur in a cluster which transfers 1 electron at a potential near that of the hydrogen couple. These potential, electron paramagnetic resonance, and absorbance properties distinguish this subclass of proteins from the “high potential iron-sulfur proteins” of Chromatium sp. and Rhodopseudomonas sp.

Proton magnetic resonance and magnetic susceptibility characterization of ferredoxin I from Bacillus polymyxa.

Magnetic susceptibility and proton magnetic resonance spectra are reported for the oxidized and reduced forms of the iron-sulfur protein Bacillus polymyxa ferredoxin I. The magnetic susceptibility of the oxidized form indicates antiferromagnetic exchange coupling between component iron atoms that is quantitatively similar to that observed for the clostridial ferredoxins and for the [(C(2)H(5))(...

Redox pathways in electron - transfer proteins : Correlations between reactivities , solvent exposure , and unpaired - spin - density distributions ( cytochromes / iron - sulfur proteins / copper proteins )

The relative reactivities toward reduction by free flavin semiquinones of cytochromes (c-type cytochromes, cytochrome b5, c'-type cytochromes) iron-sulfur proteins (high-redox-potential ferredoxins, rubredoxins, low-redoxpotential ferredoxins), and blue copper proteins (plastocyanin, azurins) are shown to correlate with calculations of the solvent exposure of the various prosthetic groups. In t...

Characterization of two soluble ferredoxins as distinct from bound iron-sulfur proteins in the photosynthetic bacterium Rhodospirillum rubrum.

In an earlier investigation (Shanmugam, K. T., Buchanan, B. B., and Arnon, D. I. (1972) Biochim. Biophys. Acta 256, 477-486) the extraction of ferredoxin from Rhodospirillum rubrum cells with the aid of a detergent (Triton X-100) and acetone revealed the existence of two types of ferredoxin (I and II) and led to the conclusion that both are membrane-bound. In the present investigation, ferredox...

Appearance of Membrane-bound Iron-Sulfur Centers and the Photosystem I Reaction Center during Greening of Barley Leaves.

Dark-grown barley (Hordeum vulgare) etioplasts were examined for their content of membrane-bound iron-sulfur centers by electron paramagnetic resonance spectroscopy at 15K. They were found to contain the high potential iron-sulfur center characterized (in the reduced state) by an electron paramagnetic resonance g value of 1.89 (the "Rieske" center) but did not contain any low potential iron-sul...

A comprehensive phylogenetic analysis of Rieske and Rieske-type iron-sulfur proteins.

The Rieske iron-sulfur center consists of a [2Fe-2S] cluster liganded to a protein via two histidine and two cysteine residues present in conserved sequences called Rieske motifs. Two protein families possessing Rieske centers have been defined. The Rieske proteins occur as subunits in the cytochrome bc1 and cytochrome b6f complexes of prokaryotes and eukaryotes or form components of archaeal e...
